Phylogenetic Diversity of Staphylococcus aureus by Random Amplification of Polymorphic DNA
نویسنده
چکیده
The polymerase chain reaction was used to obtain randomly amplified polymorphic DNA profiles for genetic fingerprinting of twelve different isolates of Staphylococcus aureus from Riyadh region, KSA using RAPD-PCR technique eleven out of forty operon primers (10-mer primers) showed polymorphism among the isolates tested generating over 65 bands, approximately 42 of which were polymorphic with sizes ranging between 300 and 3,000 bp. A numerical analysis of the genomic profiles obtained demonstrated that it was possible to differentiate the S. aureus strains. Using this technique. All the isolates were classified completely into three major groups (Sc.A, Sc.B, Sc.C) with five different subgroups. Sc. A group originated from animal host while isolates from plant and animal origins formed the Sc.B and Sc.C group. The five different subgroups of Sc. A suggested adaptation of S. aureus in different host cells. This indicates possible relationships between host origin and genetic variation among S. aureus isolates. The DNA fingerprint defined for each race of S. aureus could be useful in epidermiological studies, medical diagnosis and the identification of new strains and their origins.

Random amplified polymorphic DNA (RAPD) fingerprinting of Mycoplasma gallisepticum isolates from chickens
Ten field isolates of Mycoplasma gallisepticum (MG) from different geographical areas of Iran were analyzed by random amplification of polymorphic DNA (RAPD). RAPD analysis produced reproducible banding patterns on the basis of which various distinct amplification patterns could be detected. MG isolates compared with reference strains (S6 and Mg SS) and vaccine strain (ts-11) and demonstrated...
متن کاملMolecular typing of strains of staphylococcus aureus isolated from bone and joint lesions in lame broilers by random amplification of polymorphic DNA.
A study was conducted to investigate the relationship between the presence of localized bacterial infection and lameness in broiler fowl (Gallus gallus domesticus). Isolation of bacteria from the proximal femur, proximal tibia, and tibiotarsus from broilers with lameness revealed a probable association between lameness and the presence of Staphlylococcus aureus. Other potential pathogens, inclu...

101 isolates of Brucella spp. including the reference (n=7) and the Iranian field strains belonging to last years 1961-2003 (n=94) were biotyped and classified into 9 biotypes. Random amplified polymorphic DNA (RAPD) analysis using a ten mer-AP4 primer was generated 72 reproduciable DNA band. The typeability was 100%. The thirteen isolates were classified into five groups, each containing 2-4 b...

Cashmere is only produced from Maraz (Kurdi) goat breed in Iraqi Kurdistan Region. The objective of this study was to assess the genetic diversity of different Maraz color types, black goat and Shami breeds using random amplified polymorphic DNA (RAPD) markers. Fourteen primers were used and nine out of them were selected based on their number of bands (NB) and polymorphic characteristics. Thes...
متن کاملStudy of Genetic Diversity among Philodendron Varieties by Randomly Amplified Polymorphic DNA Markers
In the present study, Randomly Amplified Polymorphic DNA (RAPD) markers were used to analyze the genetic diversity in 20 varieties of Philodendron. The polymerase chain reaction was performed with 60 RAPD primers, out of which 21 primers showed clear amplification as well as more polymorphism. In total, 354 scorable RAPD loci with 348 polymorphic bands (98%) were observed. Percentages of polymo...
